Regulatory T Cells (Tregs) and Autoimmune Diseases

Tregs are a subset of adaptive lymphocytes. The importance of Tregs in immune regulation and brokering tolerance has been robustly demonstrated. Strategies to induce or increase Tregs have been developed to reduce immune inflammation in patients with autoimmune diseases. How to Prepare Antigen-Specific Tregs?

Tregs play a critical role in regulating the immune system by inhibiting the function of immune cells to keep immunologic self-tolerance and immune homeostasis. An acquired immunodeficiency disease (AID) will occur when the specific transcription factor forkhead box protein p3 (Foxp3) of Tregs is mutated or the CD4+CD25+ T cells are eliminated.

Antigen-specific Foxp3 Tregs can be induced by either expressing antigens in a certain tissue, such as thymic epithelial cells, or delivering antigens under sub-immunogenic conditions. In T cell antigen receptor (TCR)-transgenic mice, scientists implanted osmotic minipumps and found that when supplied 2-week, it could be used to instruct naive T cells to become Foxp3 Tregs.
